dc.description.resumoA Tripanossomíase americana, ou mais comumente conhecida, doença de Chagas (DC), é atualmente uma das mais importantes antropozoonose endêmica. É causada pelo protozoário Trypanosoma cruzi (T. cruzi). T. cruzi pode ser encontrado em diferentes locais dentro do hospedeiro vertebrado. As formas tripomastigotas sanguíneas predominam na fase aguda, bem como a infecção por amastigotas de praticamente todos os órgãos e sistemas. A carga parasitária é um fator importante que influencia o progresso do estágio agudo e, consequentemente, a doença, e está ligada a alterações cardíacas, intestinais e renais. A diabetes e a doença de Chagas foram relatadas em estudos anteriores, mas as alterações pancreáticas na doença de Chagas foram pouco estudadas e focadas em estudos funcionais de metabolismo de carboidratos e pâncreas exócrino, principalmente nas formas indeterminada e crítica da doença. Aqui estabelecemos infecção chagásica aguda e crônica em camundongos C57BL/6 machos com cepa Y e parasitemia de acesso, infiltrado inflamatório pancreático, mortalidade e histopatologia pancreática, morfometria e funcionalidade com TITG e quantificação de insulina plasmática. A infecção por T. cruzi leva ao parasitismo e consequentes alterações morfológicas e funcionais no pâncreas, de um modo dependente do inóculo. Alta carga parasitária induz a uma infecção aguda com alta parasitemia, mortalidade e menor parasitismo tecidual, com ligeiras alterações pancreáticas (estruturais e funcionais). A menor carga parasitária acarreta parasitismo pancreático acentuado, com intenso infiltrado inflamatório associado, área das ilhotas pancreáticas, associada a menor insulina plasmática e glicemia. Em camundongos na fase crônica com diferentes inóculos manteve-se de forma semelhante o parasitismo e a inflamação pancreática e menor insulina plasmática. Essas diferentes alterações pancreáticas em relação às diferentes cargas parasitárias podem ser partes importantes no amplo enigma de variações clínicas e metabólicas associadas à doença de Chagas.por
dc.description.abstractAmerican Trypanosomiasis, or more commonly known, Chagas Disease (CD), is currently one of the most important endemic anthropozoonosis. It is caused by the Trypanosoma cruzi protozoan (T. cruzi). T. cruzi can be found in different sites inside the vertebrate host; the blood trypomastigote forms predominate in the acute stage, as well as the amastigote infection is present all organ and systems. The parasite load is an important factor that influences the progress of the acute stage, and consequently, the disease, and are linked to heart, intestinal and kidney changes. Diabetes and Chagas disease was related in previous studies, but pancreatic changes in Chagas Disease were little studied and focused in functional studies of carbohydrate metabolism and exocrine pancreas, mainly in the indeterminate and critical forms of the disease. Here we did establish acute and chronic chagasic infection in male C57BL/6 mice with Y strain and access parasitaemia, pancreatic inflammatory infiltrate, mortality and pancreatic histopathology, morphometry and functionality with Intraperitoneal Glucose Tolerance Test (TITG) and plasma insulin quantification. Trypanosoma cruzi infection leads to parasitism, and consequent morphological and functional changes in the pancreas, in an inoculum dependent way. High parasite load leads to an aggressive acute infection with high parasitemia, mortality and smaller tissue parasitism, with slight pancreatic changes (structural and functional). Lower parasite load leads to marked pancreatic parasitism, with intense associated inflammatory infiltrate, reduction of the pancreatic islet area, associated with lower plasma insulin and glycemia. At chronic phase mice with different inocula similarly maintained the parasitism and the pancreatic inflammation and lower plasma insulin. These different pancreatic changes in relation to different parasite loads can be important parts in the wide variation puzzle of clinical and metabolic changes that are associated with Chagas Disease.eng
